MÖ 855 yılında Asur kralı III. Salmanassar tarafından kurulduğu zaman "Şitamrat" adını taşıyordu. Şehir, tarihi boyunca Hitit, Asur, Med, Pers, Makedon, Selevkos ve Partlar' ın idaresinde kalmıştır. Yunanlar buraya "Urima" adını vermişlerdir. Süryaniler ise "Kal'a Rhomeyta" ve "Hesna d'Romaye" adlarını kullanmışlardır. Güneydoğu Anadolu, Roma İmparatorluğu döneminde Orshoene eyaleti içinde yer almış ve kale bu eyaletteki önemli şehirlerden birisi olmuştur. 2. yüzyılda Bizanslıların eline geçince bu kez "Romaion Koyla" adını almıştır. Urfa ve çevresi Ömer döneminde fethedilmiş ve daha sonra Emevi, Abbasi, Selçuklu, Zengi ve Eyyübiler’in hâkimiyetlerinde bulunmasına rağmen Rumkale olarak bilinen yerleşim Müslüman devletlerin toprakları dışında kalmıştır. Urfa Haçlı Kontluğunu kuran Boudovin de Boulogne 1116 yılında Rumkale’yi Ermeni Prensi Gog-Vasil’in elinden aldı. Urfa kontesi Beatrice 1150 yılında Rumkale' yi, Ermeni Katolikosuna teslim etti. Rumkale, 1260 yılında İlhanlı hükümdarı Hülagu' nun orduları tarafından ele geçirildi.
